Abstract: Objective:To investigate the influence of hypoxia inducible factor-2 alpha(HIF-2α) on the expression of liver cancer stem cell marker EpCAM and CD133.Methods:Up and down-regulation of the expression of HIF-2α in HepG2 cells and Hep3B cells,respectively.Real-time PCR and Western Blot were used to detect the levels of EpCAM and CD133 mRNA and protein expression. Results:The expression of EpCAM and CD133 mRNA and protein was increased after the HIF-2α expression plasmids transfection(P<0.05),and was decreased after HIF-2α shRNA plasmids transfection(P<0.05).Conclusion:HIF-2α may play a positive role in maintaining the stemness of liver cancer cells.
