IGF-1 regulates the expression of HO-1 in gastric cancer

Abstract: Objective:To investigate the relationship between the expressions of insulin-like growth factor-1(IGF-1) and hemeoxygenase-1(HO-1) in patients with gastric cancer.To study the effects of IGF-1 on the expression of HO-1 in gastric cancer cells.Methods:We collect serum samples of patients with gastric cancer,then,ELISA was used to detect the levels of IGF-1 and HO-1 expression.Gastric cancer cells SGC7901 was used in vitro,which were intervened by IGF-1 in different concentrations.The mRNA and protein expression of HO-1 in gastric cancer cells were detected by RT-PCR and Western blot analysis.Then the IGF-1 blockers were added to the culture medium of gastric cancer cells,to observe the change of the expression of HO-1 in gastric cancer cells both before and after using blockers.Results:The expressions of IGF-1 and HO-1 in the patients with gastric cancer significantly were increased than those in the controls.The significant positive correlations were observed between the levels of IGF-1 and HO-1.Gastric cancer cells SGC7901 were cultured in vitro,and the HO-1 expression in gastric cancer cells was significantly enhanced by IGF-1 by a concentration-dependent manner.The HO-1 expression in gastric cancer cells was significantly inhibited by IGF-1 blockers.Conclusion:This experiment studies the correlation of IGF-1 and HO-1 in gastric cancer,and suggests the positive regulatory effects of IGF-1 on HO-1 expression in gastric cancer cells.Our research provides the theoretical basis for gastric cancer.
