Effects of Resveratrol on Expression of Molecules Related with mTOR Signaling Pathway in Pathological Sar Fibroblasts

Abstract: Objective In order to observe the effects of resveratrol on expression of molecules such as PI3K,A kt and mTOR related with mTOR signaling pathway in pathological scar fibroblasts.Methods The expression of PI3K,Akt and mTOR in pathological scar and normal skin tibroblasts were detected by immunofluorescence.After treated with different concentrations of resveratrol,the expression of PI3K,Akt and mTOR mRNA and protein were respectively detected by RT-PCR and Western Blot.Results Immunofluorescence showed that the expression of PI3K,Akt and mTOR were significantly enhanced in pathological scar fibroblasts,and mainly expressed in the nucleus,but no expression in normal skin fibroblasts.RT-PCR and Western Blot test resuhs showed that after treated with different concentrations of resveratrol,the mRNA and protein expression of the A kt,and mTOR were decreased in a dose dependent manner.Compared with the control group,the difference was statistically significant(P<0.05).But PI3K mRNA and protein expression was not significantly reduced,and it has no significant difference compared with the control group (P>0.05).Conclusion The mechanism of resveratrol inhibiting pathological scar fibroblast proliferation may be associated with reducing the expression of mTOR signaling pathway key molecule Akt and mTOR.
