Abstract: Objective To explore the influencing factors of amperometric method in the determination of real protein in dairy products,as well as the new methods for the rapid and accurate detection of protein content in dairy products. Methods At room temperature,put 100 ml phosphate buffered solution,10 ml 2% glutaraldehyde solution and a stirrer in a bottle,put the bottle on a magnetic stirring apparatus with the electrode of dissolved oxygen meter in it,after the base line reached a steady state,record the current value and temperature,and 10 ml certain concentration standard or sample solution were injected,read the current value after 15 min,and then to calculate the current drops. Results The glycine showed good linear relation in the range of 10 mmol / L- 50 mmol / L with the current decrease. The correlation coefficients( r) were 0. 996,and the detection limit was 0. 54 mmol / L,the average recoveries were within 81. 1%- 133. 0%,and RSDs were within 0. 306%- 3. 100%. Conclusion Under optimal conditions,the results showed that amperometric method of real protein is rapid,feasible and reliable,which can be used in the fast determination of dairy products.
