Inhibitory Effect of Schaftoside on C57BL/6 Mice with Cholesterol Gallstone Formation Induced by High-fat Diet

Abstract: Objective To study the inhibitory effect of schaftoside on cholesterol gallstone formation in experimental C57BL/6 mice induced by high-fat diet. Methods A total of 72 C57BL/6J mice were randomly divided into six groups,namely normal group,model group,ursodeoxycholic acid(UDCA)group(60 mg·kg-1),and low-,middle-,highdose schaftoside groups(in the dosage of 20,40,80 mg·kg-1 respectively),12 mice in each group. The mice were intragastrically administered for 8 weeks. Except for the normal group,the mice of other groups were fed with high-fat diet. Body weight and food intake were recorded every 7 days. We observed the stone formation with naked eyes and microscope, detected the levels of serum total cholesterol(TC), high-density lipoprotein cholesterol(HDL-C),low-density lipoprotein cholesterol(LDL-C),total bile acid(TBA),triglyceride(TG),examined bile TC,TBA,phospholipid(PL),calculated the cholesterol saturation index(CSI),and observed the liver pathological changes under light microscope. Results Compared with the model group,high-dose schaftoside group could significantly lower the rate of stone formation(33.3 %),increase serum HDL-C level and decrease TG level,reduce TC and PL level in gallbladder bile and increase TBA level, thus reducing the CSI. Compared with the model group, middle- and high-dose schaftoside groups relieved hepatic fatty degeneration in mice. Conclusion It is indicated that schaftoside can inhibit C57BL/6 mice cholesterol gallstone formation induced by high-fat diet,and the mechanism may be related with decreasing gallbladder bile CSI,increasing serum HDL-C level,and reduce liver damage resulting from hepatic lipid disorders.
