Abstract: Objective To establish the double PCR for simultaneously diagnosing Salmonella and Staphylococcus aureus. Methods According to Salmonella inv A gene and Staphylococcus aureus nuc gene sequences,the two primers were designed,and a double PCR method was established to amplify nuclear acid,which was used in the poultry farms. Results After PCR amplification,the amplified specificity bands of Salmonella 284 bp and S. aureus 484 bp were detected,while,other common pathogenic bacteria were not detected. The detection results of 1 231 broiler manure samples found that Salmonella was detected in 40 samples,with the positive rate of 2. 33%,Staphylococcus aureus was detected in 101 samples,with the positive rate of 8. 22%.Conclusion It was evident that the method based on PCR assay was highly specific and rather sensitive,and can improve the efficiency and accuracy of the detection of pathogenic bacteria in clinical stools.
