Abstract: Objective A method has been established for determination of glutathione( GSH) in health food by amino acid auto analyzer. Methods The sample was dissolved by p H = 2. 20 hydrochloric acid / citric acid buffer solution,extracted by ultrasonic extraction and filtered by the microporous membrane. After filtration,the extract solution was cleaned up by cation- exchange resin column using hydrochloric acid / citric acid buffer solution( p H 3. 20 and p H 4. 25),separated by cation exchange resin column. Chromogenic reaction between glutathione and ninhydrin was detected at wave length of 570 nm; The column temperature was 44℃ and flow rate was 35 ml / h; the reaction temperature was 135℃ and injection volume was 20 μL. Results The linear range of the method was good under the concentration of the control sample between 1. 0 μg / ml and 100. 0 μg / ml; linear regression equation was y = 19. 974 x + 12. 379 with correlation coefficient( r) > 0. 999; the limit of quantification was0. 63 μg / ml,the detection limit was 0. 19 μg / ml,the average recovery rate was 102. 1%,and the relative standard deviation( RSD) of peak area was 1. 8%. Conclution The method is simple,rapid,sensitive and accurate,and it can be used for the determination of GSH in health food.
