Abstract: Southeastern China is one of the high prevalence areas for nasopharyngeal cancer(NPC) in the world. There is a strong need to investigate the mechanism of NPC metastasis. It has theoretic importance in medicine and great value in clinical application to explore the correlation between LMP-1 cancer protein and metastatic mechanism by utilizing optical visualization techniques for the early prediction of risk of metastasis, possible preventive intervention and improvement in survival rate. This review summarizes recent research development and trend in applying high resolution optical imaging and fluorescence correlation spectroscopy technology, including study on specific target of the LMP-1 protein. In combination with high sensitivity fluorescent probes, high resolution optical imaging of lipid rafts can be obtained for the optical characterization of dynamic aggregation properties of LMP-1 in lipid rafts, which can be used for the visualization and semi-quantified prediction of the trend of metastasis. It may provide a new direction and theoretic support for a significant improvement of the cure rate in NPC treatment.
