Performance evaluation of white blood cells and nucleated red blood cells counting via Sysmex XN-2000 and XE-5000 automatic hematology system

Abstract: Objective To conduct performance evaluation of nucleated red blood cells(NRBC) and white blood cells(WBC) test in peripheral blood via Sysmex XN-2000 and XE-5000 automatic hematology system. Methods A total of 262 subjects were enrolled,which included 151 males and 111 females,aged 5-76 years old with mean age of 42 years old. All of them were detected by Sysmex XN-2000 and XE-5000 automatic hematology system,respectively. The counting results of NRBC and WBC between 2instruments were compared. Meanwhile,microscope counting was used as reference method. Results The sensitivity and negative predictive value of NRBC with XN-2000 was 100 %,specificity was 97.78 % and positive predictive value was 87.81%. There was no statistical difference between the results of NRBC counting and percentage from XN-2000 and microscope method(P=0.223 1,0.418 3,respectively),and showed significant positive correlation(r=0.974,0.943,respectively). The statistical difference of WBC counting,neutrophils and lymphocytes percentage,monocyte percentage existed between the results from XN-2000 and XE-5000test(P=0.000 8,0.004 0,0.001 0,0.003 0,respectively),while no statistical differences were observed when compared the results with counterparts of correction results from XE-5000(P=0.251 1,0.752 3,0.224 7,0.414 5,respectively). Conclusion It is demonstrated that Sysmex XN-2000 automatic hematology system can detect NRBC sensitively and accurately,and correct WBC counting results automatically,which reduce the reviewing rate and shorten the specimen detection time.
