Abstract: Objective To investigate the effects of arensic trioxide(ATO) on human hepatocarcinoma cell lines HepG2 and Huh-7,and explore its mechanisms. Methods HepG2 and Huh-7 cells were cultured in vivo and incubated with different concentrations of ATO(0,2,4 μmol/L). After treatment,cell morphology was observed with inverted microscope and cell proliferation was determined by MTT assay,cell apoptosis by flow cytometry with annexin V/propidium iodided staining,and Bcl-2111(Bim) was detected by Western-blot. Results The ATO with different concentrations(0,2,4 μmol/L) showed different degrees inhibitory effects on growth of 2 cells and concentration-dependent,which was no significant difference between HepG2 and Huh-7 cells. The ATO showed obvious effect on promoting apoptosis,and the Bim of 2 cell lines showed high expression after ATO treatment.Conclusion It is demonstrated that ATO could be induced apoptosis effect on HepG2 and Huh-7 cells.
