Dexmedetomidine intervention effects on the expression of IL-10 in pulmonary tissue of rats with sepsis-associated acute lung injury

Abstract: Objective To study the regulation of IL-10 change in the lung tissue of rats with sepsis-associated acute lung injury(ALI),and to investigate the dexmedetomidine protective effect on lung of rats with sepsis-associated ALI. Methods 75 male Sprague-Dawley(SD)rats were divided into sham operation group,sepsis model group and dexmedetomidine group,25 rats in each group. The lung wet/dry(W/D)weight ratio was detected,and The expression of IL-10 in lung tissue of rats in each group were detected by immunohistochemistry method and Western blot method. Results Compared with sham operation group,the W/D ratio of rats in ALI group was higher(P<0.05),however,the W/D ratio of rats in dexmedetomidin group was decreased significantly after dexmedetomidin treatment(P<0.05). Compared with sham operation group,the mean optical density(MOD)value of IL-10 positive expressions in lung tissue of rats in sepsis model group was lower. Compared with sepsis model group,the MOD value of IL-10 positive expressions in lung tissue of rats in dexmedetomidin group was increased significantly(P<0.05). Conclusion Dexmedetomidine could attenuate lung inflammatory reaction in septic rats by up-regulating the expression of IL-10.
