The Expression of angiopoietin-2 on retinopathy of prematurity model

Abstract: Objective To investigate the expression of angiopoietin-2(Ang-2)in the retina of hyperoxia-induced model of retinopathy and to evaluate the effect of Ang-2 in retinal neovascularization. Methods C57BL/6J mice were put into the environment of high oxygen to establish models of ischemic retinopathy. Twenty mice were randomly divided into hyperoxic and normoxic control groups. The hyperoxic group mice were put into closed container where oxygen concentration was 75±2% for 5 days,then moved into normal air. The normoxic group mice were raised in normal air. Mice were sacrificed in postnatal day 17,the expression of Ang-2 was examined in the mice retina using immunohistochemistry and reverse transcription-polymerase chain reaction.Results The neovascular endotheliocyte was higher in hyperoxic group(25.68±2.61)than which in normoxic group(1.45±1.01)(P<0.05). The m RNA expression of Ang-2 was higher in hyperoxic group(3.01±1.11)than which in normoxic group(1.47±0.53)(P<0.05). The optical density value of Ang-2 was higher in hyperoxic group(121.25±8.61)than which in normoxic group(75.45±7.87)(P<0.05). Conclusion The retinal neovascularization and the Ang-2 expression were increased in hyperoxic group,suggesting that Ang-2 may related to retinal neovascularization.
